I don't thing a rockwell test would give you a very accurate reading considering how soft human tissue is vs any material its used on, (not to mention no one is going to volunteer to have a 120degree diamond cone or 10mm ball pressed into their penis). I could see maybe something like a brinell hardness test accept instead of using 10,000psi, perhaps something like 50 psi...
